Do not Grieve the Spirit

In Matt. 28:19, Jesus instructed His disciples to “make disciples of all the nations” and to be witnesses “to the end of the earth” (Acts 1:8). But in Acts 16:6, why did the Holy Spirit forbid Paul and his companions to preach the word in Asia?
Rom. 8:26 says that the Holy Spirit intercedes for us to God “with groanings which cannot be uttered.” But 1 Tim. 2:5 says that “there is one God and one Mediator between God and men, the Man Christ Jesus.” How can Jesus be the only mediator when the Holy Spirit is also seen as a mediator?
